Human Eukaryotic Translation Initiation Factor 6 ELISA Kit

Sandwich ELISA kit from Antibodies.com for the quantitative in vitro determination of human Eukaryotic Translation Initiation Factor 6 (eIF-6) in tissue homogenates, cell lysates or other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips), a biotinylated anti-eIF-6 detection antibody and HRP-avidin with TMB substrate, read at 450 nm. Reported sensitivity is 0.123 ng/ml and the working assay range is 0.312–20 ng/ml, with a total assay time of 4h 30m.

The colourimetric, quantitative sandwich immunoassay has been evaluated in serum, EDTA plasma and heparin plasma. Average recovery is 80%–102% (91%, n=5) in serum, 81%–100% (90%, n=5) in EDTA plasma and 80%–89% (84%, n=5) in heparin plasma. Linearity at 1:2, 1:4, 1:8 and 1:16 dilutions is 87–91%, 87–107%, 74–101% and 92–97% respectively for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components include a pre-coated 96-well microplate, lyophilised standard (2 vials), Detection Solution A (120 μl) and Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips), stored at +4°C or −20°C as instructed.

Required materials not supplied include a microplate reader capable of measuring absorbance at 450 nm ± 10 nm, a multichannel pipette reservoir or automated microplate washer, a 37°C incubator, multi- and single-channel pipettes with sterile tips, deionised or distilled water, absorbent paper towels, test tubes and Eppendorf tubes, and computer software capable of generating logarithmic functions.
