The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for Elongation Factor 1-gamma that has been pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added into the wells, and bound analyte is detected using a Biotinylated Anti-Elongation Factor 1-gamma Antibody followed by HRP-Avidin conjugate.

TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Elongation Factor 1-gamma captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader against the standard curve. The assay reports a product range of 0.312-20 ng/ml with a sensitivity of 0.114 ng/ml, and the total assay time is 4h 30m.

Validated sample recovery averages are 91% for serum (range 83% - 100%), 96% for EDTA plasma (range 90% - 103%), and 95% for heparin plasma (range 89% - 102%). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
