Human Erythropoietin ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Erythropoietin (EPO) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses a pre-coated microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where EPO is captured by an immobilised antibody, followed by incubation with a Biotinylated Anti-EPO Antibody and an HRP-Avidin conjugate. TMB substrate is then added, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution; the density of yellow is proportional to the amount of EPO captured. Absorbance is read at 450 nm in a colorimetric microplate reader, and the total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed.

Reported performance characteristics include a measuring range of 31.2-2000 pg/ml and a sensitivity of 12.7 pg/ml. Spike-and-recovery values were serum (n=5) 80%-102% (average 91%), EDTA plasma (n=5) 81%-100% (average 90%), and heparin plasma (n=5) 80%-89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions was 87-91%, 87-107%, 74-101% and 92-97% respectively for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).
