This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human CXCL5, also known as Epithelial Neutrophil Activating Peptide 78. The assay is validated for use with human serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, with reactivity restricted to human samples. A CXCL5-specific capture antibody is pre-coated onto a 96-well microtiter plate; standards and samples are added, the captured analyte is detected using a biotinylated anti-CXCL5 antibody, and an HRP-Avidin conjugate is introduced. TMB substrate produces a colour change proportional to analyte concentration, which is read at 450 nm on a colorimetric microplate reader.

The standard curve covers 0.156–10 ng/ml with a sensitivity of 0.063 ng/ml. Spike-and-recovery was assessed across five replicates per matrix, yielding average recoveries of 91% for serum (range 83–100%), 95% for EDTA plasma (range 91–100%) and 96% for heparin plasma (range 91–101%). Total assay time is approximately 4 hours 30 minutes from sample addition to final read, and the plate should be read using a colourmetric microplate reader at 450 nm.
