Human Eosinophil Peroxidase ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Eosinophil Peroxidase (EPX) in serum, plasma, tissue homogenates and other biological fluids.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, where EPX is captured by the immobilised antibody. After washing, a Biotinylated Anti-EPX Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of EPX captured, and the concentration is calculated by reading absorbance at 450 nm in a microplate reader against the standard curve.

Sensitivity is reported as <0.51 ng/ml with an assay range of 1.25-80 ng/ml, and the total assay time is 4 hours 30 minutes. Validation data show average recovery of 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%) and 84% in heparin plasma (range 80-89%).
