Human Eomesodermin ELISA Kit

Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit designed for the in vitro quantitative determination of human TBR2/Eomes (Eomesodermin) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using a pre-coated 96-well microplate (12 x 8 well strips) to which standards and test samples are added. Following incubation and washing, a Biotinylated Anti-TBR2/Eomes Antibody binds the captured analyte, then HRP-Avidin conjugate is added. TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of TBR2/Eomes captured, and concentration is calculated by reading O.D. absorbance at 450 nm.

The kit has a reported sensitivity of 0.115 ng/ml and a standard curve range of 0.312–20 ng/ml. Total assay time is 4 hours 30 minutes, and detection is performed with a colorimetric microplate reader. Supplied components include the pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Storage temperatures differ by component: most items are kept at -20°C, while wash buffer, TMB substrate, and stop solution are stored at +4°C.

Spike-and-recovery performance (n=5) averaged 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Linearity-of-dilution data is reported for serum, EDTA plasma, and heparin plasma samples at 1:2, 1:4, 1:8, and 1:16 dilutions.
