A sandwich Enzyme-Linked Immunosorbent Assay designed for the in vitro quantitative determination of human Endoplasmic Reticulum To Nucleus Signalling 1 (IRE1) in serum, plasma, tissue homogenates or other biological fluids. The kit is supplied by Antibodies.com under their Assays and Kits category and has been validated for reactivity with human specimens.

Reported performance characteristics include a measuring range of 0.156–10 ng/ml and a sensitivity of <0.071 ng/ml. The total assay time is approximately 4 hours and 30 minutes. Average recoveries in undiluted samples were 91% for serum, 90% for EDTA plasma and 84% for heparin plasma (n=5 per matrix), with linear dilution recoveries reported across 1:2, 1:4, 1:8 and 1:16 dilutions. Detection is colorimetric and the absorbance is read at 450 nm in a microplate reader.

The assay uses a pre-coated microplate arranged as 12 x 8 well strips. An antibody specific for IRE1 is immobilised on the plate; standards and test samples are added and any IRE1 present is captured. After washing, a biotinylated anti-IRE1 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then added and the enzymatic catalysis produces a blue colour, which turns yellow upon addition of the acidic stop solution; the intensity of the yellow colour is proportional to the amount of IRE1 captured. Kit components include the pre-coated plate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers. Components should be stored at +4°C or –20°C as instructed and the kit should not be used past its expiration date.
