Human Endonuclease G, Mitochondrial ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com designed for the in vitro quantitative determination of human mitochondrial Endonuclease G (Endo G) in tissue homogenates, cell lysates, and other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for Endo G, with standards and samples incubated in the wells before washing and incubation with a biotinylated anti-Endo G antibody. An HRP-avidin conjugate is then added, and the enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The optical density is read at 450 nm in a microplate reader.

The kit has a reported sensitivity of <0.062 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery was evaluated in serum (average 91%, range 80%–102%), EDTA plasma (average 90%, range 81%–100%), and heparin plasma (average 84%, range 80%–89%) across five samples per matrix. Linearity was assessed at dilutions of 1:2, 1:4, 1:8, and 1:16 in the same matrices. Components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and plate sealers, with storage conditions of +4°C or -20°C as specified per component.
