Human Endo Beta-N-Acetylglucosaminidase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com enables the in vitro quantitative determination of human ENGASE (Endo Beta-N-Acetylglucosaminidase) in tissue homogenates, cell lysates and other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using a pre-coated microplate to which an antibody specific for ENGASE has been immobilised. Standards and test samples are added to the wells; ENGASE present in each sample is captured by the immobilised antibody and detected using a Biotinylated Anti-ENGASE Antibody followed by an HRP-Avidin conjugate. A TMB substrate solution is used to visualise the reaction, producing a blue product that turns yellow upon addition of stop solution. The concentration of ENGASE is then calculated from the absorbance read at 450 nm in a microplate reader, by reference to a standard curve.

The reported sensitivity is <0.125 ng/ml and the assay range is 0.312–20 ng/ml. Total assay time is approximately 4 hours 30 minutes, and the assay is read on a colorimetric microplate reader. Kit components are stored at +4°C or −20°C as instructed, and the kit is described as suitable for cell lysates and tissue homogenates. Reported average recovery values across the tested matrices are 91% for serum (range 80–102%), 91% for EDTA plasma (range 81–101%) and 84% for heparin plasma (range 80–89%), each from n=5 samples.

Reactivity is confirmed for human samples, and the kit is intended for the quantitative measurement of the ENGASE target using a pre-coated microplate format with colorimetric detection.
