Human Elastase 2, Neutrophil ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables in vitro quantitative determination of human Neutrophil Elastase in serum, plasma, tissue homogenates, cell lysates and other biological fluids. The platform is a 96-well microplate pre-coated with an antibody specific for Neutrophil Elastase; standards and samples are added and bound analyte is detected with a biotinylated anti-Neutrophil Elastase antibody, followed by HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that turns yellow upon addition of acidic stop solution, and the intensity of the yellow colour is read by absorbance at 450 nm on a colorimetric microplate reader and quantified against a standard curve.

Reported performance characteristics include a product range of 0.156–10 ng/ml and a sensitivity of 0.062 ng/ml, with a total assay time of 4 hours 30 minutes. In recovery testing (n=5), average values were 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed in the protocol manual and should not be used past the stated expiration date.

The assay is reactive with human samples and is intended for the quantitative measurement of Neutrophil Elastase across the listed biological fluid and tissue preparations.
