Human Elastase 1, Pancreatic ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay designed for the in vitro quantitative determination of human Elastase 1 in serum, plasma, tissue homogenates, cell lysates or other biological fluids, this kit is offered by Antibodies.com under their Assays and Kits product category. It is reactive with human samples and uses the sandwich enzyme immunoassay technique, in which an antibody specific for Elastase 1 is pre-coated onto a microplate. Standards and test samples are added; captured Elastase 1 is then bound by a Biotinylated Anti-Elastase 1 Antibody. An HRP-Avidin conjugate and TMB substrate produce a yellow colour whose intensity is read at 450 nm on a colorimetric microplate reader.

The quantitative measuring range is 62.5-4000 pg/ml with a sensitivity of <23.6 pg/ml, and the total assay time is 4h 30m. Kit components are stored at +4°C or -20°C as instructed.

Validation data show spike-and-recovery ranges in serum (n=5) of 80%-102% (91% average), in EDTA plasma (n=5) of 81%-100% (90% average), and in heparin plasma (n=5) of 80%-89% (84% average). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 gave recoveries of 87-91%, 87-107%, 74-101% and 92-97% for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).
