Human Ectonucleotide Pyrophosphatase/Phosphodiesterase 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human ENPP1/PC1 in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) and employs a biotinylated anti-ENPP1/PC1 detection antibody with HRP-avidin and TMB substrate for colorimetric readout measured at 450 nm. Standards and samples are added to the antibody-coated wells and bound analyte is detected sequentially, producing a yellow product whose absorbance is proportional to ENPP1/PC1 concentration when read against a standard curve.

Sensitivity is <0.062 ng/ml with an assay range of 0.156-10 ng/ml and a total assay time of 4h 30m. Average recovery in neat samples (n=5) was 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%). Dilution linearity was demonstrated across 1:2 to 1:16 dilutions in serum (87-91%, 87-107%, 74-101%, 92-97%), EDTA plasma (90-105%, 84-101%, 90-101%, 79-108%) and heparin plasma (84-95%, 92-105%, 82-105%, 89-91%).
