Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Dystrobrevin Beta (DTN-B) in tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for DTN-B pre-coated onto a microplate. Standards and test samples are added to the wells, where DTN-B present in each sample is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-DTN-B Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of DTN-B captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a stated sensitivity of <0.055 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Detection is colourimetric on a pre-coated microplate read with a standard colorimetric microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The protocol is designed to be easy to follow.

Alongside tissue homogenates, the kit is validated for serum, EDTA plasma, and heparin plasma samples, with reported average recovery values of 91% for serum, 90% for EDTA plasma, and 84% for heparin plasma.
