The assay is built around a pre-coated 96-well microplate formatted as 12 x 8 well strips, paired with a biotinylated anti-DNHD1 detection antibody and an HRP-avidin/TMB substrate system. Signal is read as optical density at 450 nm on a colorimetric microplate reader. The standard curve covers an assay range of 0.312-20 ng/ml with a sensitivity of 0.112 ng/ml, and the complete protocol takes approximately 4 hours 30 minutes.
