A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human DYRK1A (Dual Specificity Tyrosine Phosphorylation Regulated Kinase 1A). The kit is suitable for tissue homogenates, serum, plasma (EDTA or heparin), cell culture supernatants, cell lysates, and other biological fluids. It uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a biotinylated anti-DYRK1A detection antibody, an HRP-avidin conjugate, and TMB substrate. The enzymatic reaction is read by absorbance at 450 nm in a colourimetric microplate reader, and the assay is quantitative by reference to a standard curve.

The standard curve range is 0.312–20 ng/ml with a sensitivity of <0.117 ng/ml, and the total assay time is 4h 30m. Reported recovery values (n=5) are 83–100% for serum (average 91%), 90–98% for EDTA plasma (average 94%), and 89–102% for heparin plasma (average 95%). Linearity across 1:2, 1:4, 1:8 and 1:16 dilutions has been reported as 83–110%, 79–100%, 90–98% and 82–108% for serum; 90–98%, 90–105%, 73–109% and 87–91% for EDTA plasma; and 88–103%, 77–105%, 95–98% and 87–110% for heparin plasma. A standard dilution series prepared from a 20 ng/ml stock gives 10, 5, 2.5, 1.25, 0.625 and 0.313 ng/ml.

Kit reagents are stored at +4°C or -20°C as instructed, and the product is for research use only; it is not intended for diagnostic or therapeutic applications. The Stop Solution is an acid and TMB Substrate is light-sensitive, so protective equipment and avoidance of prolonged substrate exposure are recommended.
