Description

From Antibodies.com, this colourmetric Sandwich ELISA kit is designed for the in vitro quantitative determination of human Dual Specificity Phosphatase 9 (DUSP9) in tissue homogenates or other biological fluids. The kit is intended for use with human samples and is performed on a pre-coated microplate platform using a sandwich enzyme immunoassay format.

The microplate is pre-coated with an antibody specific for DUSP9. Standards and test samples are added to the wells and DUSP9 present is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-DUSP9 Antibody binds the captured target, followed by the addition of an HRP-Avidin conjugate. TMB substrate solution is then used to visualize the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of DUSP9 captured per well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader against the standard curve.

According to the manufacturer, the assay range is 0.312–20 ng/ml with a sensitivity of <0.117 ng/ml, and a total assay time of 4h 30m. Reported average recovery values are 91% for serum (n=5, range 80–102%), 90% for EDTA plasma (n=5, range 81–100%), and 84% for Heparin plasma (n=5, range 80–89%). Kit components should be stored at +4°C or -20°C as instructed, and not used past the expiration date.
