Human Dual Oxidase 2 ELISA Kit

From Antibodies.com, this sandwich ELISA is intended for the in vitro quantitative determination of human Dual Oxidase 2 (DUOX2) in tissue homogenates and other biological fluids. The procedure uses a pre-coated microplate bearing an immobilised anti-DUOX2 antibody; standards and samples are added, followed by a biotinylated anti-DUOX2 detection antibody and an HRP-avidin conjugate. TMB substrate is then added, the reaction stopped with an acidic solution, and the absorbance read at 450 nm in a microplate reader to calculate DUOX2 concentration from a standard curve.

The reported sensitivity is 0.069 ng/ml and the assay range is 0.156–10 ng/ml. The protocol is described as suitable for serum, EDTA plasma, heparin plasma, tissue homogenates, cell lysates and cell culture supernatants. Reported average recovery is 91% in serum (range 80–102%), 90% in EDTA plasma (81–100%) and 84% in heparin plasma (80–89%). Linearity is reported across dilutions from 1:2 to 1:16, with serum ranging 74–107%, EDTA plasma 79–108% and heparin plasma 82–105% across the dilution series. Total assay time is 4 h 30 min.

Components are stored at +4 °C or −20 °C as instructed and the product is for research use only. Required materials not supplied include a microplate reader capable of measuring absorbance at 450 nm, a 37 °C incubator, deionised or distilled water and appropriate pipettes.
