Human Doublecortin ELISA Kit

A sandwich ELISA from Antibodies.com for the in vitro quantitative determination of human Doublecortin in tissue homogenates, cell lysates, and other biological fluids. The assay uses a 96-well microplate pre-coated with a capture antibody specific for Doublecortin. Standards and samples are incubated in the wells and captured Doublecortin is detected using a biotinylated anti-Doublecortin antibody followed by an HRP-avidin conjugate. After incubation and addition of TMB substrate, the enzymatic reaction is stopped with an acidic solution, producing a yellow colour whose intensity is proportional to the amount of Doublecortin bound in each well. Absorbance is read at 450 nm in a colorimetric microplate reader and concentrations are calculated against a standard curve.

Performance characteristics include a working range of 0.156–10 ng/mL, a sensitivity of <0.058 ng/mL, and a total assay time of approximately 4 hours 30 minutes. In spike-and-recovery testing (n=5 per matrix), serum samples averaged 91% recovery (range 80–102%), EDTA plasma averaged 90% (range 81–100%), and heparin plasma averaged 84% (range 80–89%). Linearity was also evaluated across 1:2, 1:4, 1:8, and 1:16 dilutions in serum, EDTA plasma, and heparin plasma. Kit components are stored at +4°C or –20°C as instructed in the protocol, and the kit should not be used past its expiration date.
