Human Dopamine Beta Hydroxylase ELISA Kit

Sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Dopamine beta Hydroxylase in serum, plasma, tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for Dopamine beta Hydroxylase is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells and the captured Dopamine beta Hydroxylase is detected using a Biotinylated Anti-Dopamine beta Hydroxylase Antibody followed by an HRP-Avidin conjugate. A TMB substrate is then added to visualise the HRP enzymatic reaction, producing a blue product that turns yellow on addition of acidic stop solution. The density of the yellow colour is proportional to the amount of analyte captured, and the concentration is calculated from the O.D. absorbance read at 450 nm in a colorimetric microplate reader.

Reported performance includes a product range of 3.12–200 ng/ml and a sensitivity of <1.24 ng/ml. Spike-and-recovery across sample matrices averages 91% for serum (range 83%–99%, n=5), 89% for EDTA plasma (range 80%–99%, n=5) and 90% for heparin plasma (range 82%–99%, n=5). Total assay time is 4 h 30 min. The kit is supplied with a pre-coated 96-well microplate, lyophilised standard (2 vials), Detection Solutions A and B (120 µl each), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers. Components are stored at +4 °C or –20 °C as instructed and the kit should not be used past its expiration date.
