Human Dopachrome Tautomerase ELISA Kit

From Antibodies.com, this assay kit is intended for the in vitro quantitative determination of Dopachrome Tautomerase (TRP2/DCT) in serum, plasma, tissue homogenates and other biological fluids. It is a sandwich ELISA with reactivity for human samples.

The colorimetric assay runs in 4h 30m, with absorbance read at 450 nm on a microplate reader. The stated range is 0.312-20 ng/ml with a sensitivity of 0.125 ng/ml. Reported recovery values include serum (n=5) at 80-102% (average 91%), EDTA plasma (n=5) at 81-100% (average 90%) and heparin plasma (n=5) at 80-89% (average 84%), with linearity-of-dilution data given for 1:2, 1:4, 1:8 and 1:16 dilutions of each sample type.

Capture antibody specific for TRP2/DCT is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Samples and standards are added, washed, then incubated with a biotinylated anti-TRP2/DCT detection antibody followed by HRP-Avidin conjugate. TMB substrate produces a blue product that becomes yellow after acidic stop solution; absorbance intensity at 450 nm is proportional to TRP2/DCT concentration, quantified against a standard curve. Kit contents include the pre-coated plate, lyophilized standard, detection reagents, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or -20°C as instructed.
