Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com, designed for the in vitro quantitative determination of human DPP10 (Dipeptidyl Peptidase 10) in serum, plasma, tissue homogenates or other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. Standards and test samples are added to the wells of a pre-coated 96-well microtiter plate bearing an antibody specific for DPP10; after incubation and washing, a Biotinylated Anti-DPP10 Antibody binds the captured DPP10. An HRP-Avidin conjugate is then added, and following a further incubation and washing step, TMB substrate solution is used to visualise the HRP enzymatic reaction. The catalysed product is blue and turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of DPP10 captured per well. DPP10 concentration is calculated by reading the O.D. absorbance at 450nm on a microplate reader and referring to the standard curve.

Reported performance characteristics include an assay range of 0.312-20 ng/ml, a sensitivity of 0.115 ng/ml, an assay time of 4h 30m, and colourmetric detection read on a colorimetric microplate reader. Spike-and-recovery averages (n=5) were 91% for serum (range 83-100%), 96% for EDTA plasma (range 90-103%) and 95% for heparin plasma (range 89-102%). Dilution linearity (n=5) across 1:2, 1:4, 1:8 and 1:16 dilutions was 83-110%, 79-100%, 90-98% and 82-108% for serum; 90-98%, 90-105%, 73-109% and 87-91% for EDTA plasma; and 88-103%, 77-105%, 95-98% and 87-110% for heparin plasma. Kit components are stored at +4°C or -20°C as instructed.
