Human Dihydropyrimidine Dehydrogenase ELISA Kit

From Antibodies.com, this human Dihydropyrimidine Dehydrogenase (DPD) sandwich ELISA kit is designed for the in vitro quantitative determination of DPD in serum, plasma, tissue homogenates and other biological fluids. The assay is performed on a pre-coated microplate (12 x 8 well strips) bearing an antibody specific for DPD.

Standards and samples are added to the wells, where DPD is captured by the immobilised antibody. After washing, a biotinylated anti-DPD antibody binds the captured target, followed by addition of an HRP-avidin conjugate. TMB substrate is added to visualise the HRP reaction, producing a blue product that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader. Reported sensitivity is <0.65 ng/ml, with a product range of 1.56-100 ng/ml, and a total assay time of 4h 30m.

Recovery studies returned an average of 91% (80-102%, n=5) for serum, 90% (81-100%) for EDTA plasma and 84% (80-89%) for heparin plasma. Dilution linearity is demonstrated from 1:2 through 1:16 across all three sample matrices. The kit includes a pre-coated 96 well microplate, lyophilised standard (2 vials), detection solutions A and B (120 µl each), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and 5 adhesive plate sealers. Components are stored at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.
