Human Dickkopf Related Protein 2 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com is designed for the in vitro quantitative determination of human Dickkopf Related Protein 2 (DKK2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate in a 12 x 8 well strip format and employs the sandwich enzyme immunoassay technique, in which DKK2 in standards and samples is captured by an immobilised antibody and detected using a Biotinylated Anti-DKK2 Antibody followed by an HRP-Avidin conjugate.

Signal is developed with TMB substrate and read on a colorimetric microplate reader at 450 nm, with the resulting yellow colour intensity proportional to DKK2 concentration. According to the manufacturer's specifications, the assay has a sensitivity of 34 pg/ml, a quantitative range of 78.1-5000 pg/ml, and a total run time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Validation data supplied by the manufacturer report the following spiked recovery ranges (n=5 per sample type): serum 80%-102% with an average of 91%, EDTA plasma 81%-100% averaging 90%, and heparin plasma 80%-89% averaging 84%. Dilution linearity was evaluated at 1:2, 1:4, 1:8 and 1:16 dilutions, yielding serum recoveries of 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma recoveries of 90-105%, 84-101%, 90-101% and 79-108%; and heparin plasma recoveries of 84-95%, 92-105%, 82-105% and 89-91%.
