Human Diaphanous Homolog 1 ELISA Kit

A sandwich ELISA from Antibodies.com for the in vitro quantitative determination of human Diaphanous Homolog 1 (DIAPH1) in tissue homogenates and other biological fluids. The assay is run on a pre-coated microplate in a 12 x 8 well strip configuration with colourmetric detection read at 450 nm in a microplate reader. An antibody specific for DIAPH1 is immobilised on the plate; standards and samples are added, followed by a biotinylated anti-DIAPH1 antibody, an HRP-avidin conjugate, and TMB substrate. The resulting yellow product (formed after addition of an acidic stop solution) is proportional to the amount of DIAPH1 captured in each well. The reported measuring range is 0.156–10 ng/ml, with a sensitivity of <0.051 ng/ml and a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Spike-and-recovery performance was evaluated in serum (n=5; range 80%–102%, average 91%), EDTA plasma (n=5; range 81%–101%, average 91%) and heparin plasma (n=5; range 80%–89%, average 84%). Dilution linearity was demonstrated across 1:2 to 1:16 dilutions in serum, EDTA plasma and heparin plasma. The kit comprises a pre-coated 96-well microplate (12 x 8 well strips), a lyophilised standard, detection solutions A and B, a 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, and is suitable for tissue homogenates.
