A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of human DGAT2 in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated 96-well microplate and the sandwich enzyme immunoassay technique, in which an antibody specific for DGAT2 is immobilised on the plate, the target is captured, and detection is achieved with a biotinylated anti-DGAT2 antibody followed by an HRP-Avidin conjugate and TMB substrate. The colour change is read at 450 nm in a microplate reader and DGAT2 concentration is calculated against a standard curve.

The reported sensitivity is 0.071 ng/ml and the assay range is 0.156–10 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Recovery has been evaluated in serum, EDTA plasma and heparin plasma (n=5), with average values of 91%, 90% and 84% respectively. Linearity has also been examined at dilutions of 1:2, 1:4, 1:8 and 1:16 across the same sample types. The product is intended for research use only and is not for diagnostic or therapeutic applications.

Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer concentrate, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, with storage conditions of +4°C or -20°C as indicated for each component. Reagents should be brought to room temperature before use, and the TMB substrate equilibrated for 30 minutes at 37°C prior to addition.
