Human Desmocollin 3 ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit is designed for the in vitro quantitative determination of human Desmocollin 3 in serum, plasma, tissue homogenates and other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips), where an antibody specific for Desmocollin 3 is immobilised to capture the analyte from standards and test samples.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.062 ng/ml. Detection is colorimetric: captured Desmocollin 3 is bound by a Biotinylated Anti-Desmocollin 3 Antibody, followed by an HRP-Avidin conjugate and TMB substrate, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and concentration is calculated against a standard curve. Total assay time is approximately 4 hours 30 minutes.

Kit components include the pre-coated microplate, 2 vials of lyophilised standard, 120 μl each of Detection Solution A and Detection Solution B, 20 ml of 30X Wash Buffer, 45 ml of Sample Dilution Buffer, 9 ml of TMB Substrate, 6 ml of Stop Solution and 5 adhesive plate sealers, to be stored at +4°C or -20°C as instructed. Manufacturer-reported recovery averages are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%).
