The assay uses a pre-coated capture antibody; standards and samples are added, followed by a biotin-conjugated detection antibody and HRP-conjugated Avidin. TMB substrate produces a colour change measured spectrophotometrically at 450 nm, with target concentration calculated by comparison to a standard curve.

Quoted performance figures are a detection range of 125 - 8000 pg/ml and a sensitivity of 47 pg/ml. The capture antibody is raised in rabbit (IgG isotype).
