The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for DMBT1 pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells where DMBT1 is captured by the immobilized antibody, followed by incubation with a Biotinylated Anti-DMBT1 Antibody and then an HRP-Avidin conjugate. A TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of an acidic stop solution; the density of yellow is proportional to the amount of DMBT1 captured in each well.

Concentration is calculated by reading the O.D. absorbance at 450nm on a colorimetric microplate reader and referring to the standard curve. The kit has a stated sensitivity of 0.058 ng/ml and a working range of 0.156–10 ng/ml. Total assay time is approximately 4 hours 30 minutes. Sample handling and recovery have been evaluated for serum, EDTA plasma and heparin plasma.
