Human Deiodinase, Iodothyronine, Type II ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human DIO2 (Deiodinase, Iodothyronine, Type II) in tissue homogenates, cell lysates or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for DIO2. Standards and test samples are added, and DIO2 present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-DIO2 Antibody binds the captured analyte, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; the absorbance is read at 450 nm in a colorimetric microplate reader.

Quantitative performance characteristics include an assay range of 0.156–10 ng/ml and a sensitivity of 0.053 ng/ml, with a total assay time of 4h 30m. Sample recovery (n=5) has been validated in serum (average 91%, range 80–102%), EDTA plasma (average 90%, range 81–100%), and heparin plasma (average 84%, range 80–89%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across these same sample types, with reported ranges spanning 74–108% across all tested conditions.
