From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) enables the in vitro quantitative determination of human beta 2 Defensin/BD-2 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for beta 2 Defensin/BD-2. Standards and samples are added to the wells; following incubation and washing, a Biotinylated Anti-beta 2 Defensin/BD-2 Antibody is applied, followed by an HRP-Avidin conjugate. The reaction is visualised with TMB substrate, producing a blue colour that turns yellow after addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the concentration of beta 2 Defensin/BD-2 is calculated by reference to a standard curve.

The standard curve covers a product range of 46.8–3000 pg/ml, with a sensitivity of <13.55 pg/ml, and the total assay time is 4 hours 30 minutes. The kit is supplied as a pre-coated 96-well microplate (12 × 8 well strips) together with lyophilised standard, detection solutions A and B, wash buffer (30×), sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers. Storage requirements vary by component between +4°C and −20°C, and the kit should not be used past its expiration date.

Validation data report average recovery ranges of 83–99% in serum (mean 91%), 80–99% in EDTA plasma (mean 89%) and 82–99% in heparin plasma (mean 90%). Linearity was demonstrated at dilutions of 1:2, 1:4, 1:8 and 1:16 across the same sample types, with values typically falling within 74–110% across the dilution series.
