Human Defensin Alpha 5, Paneth Cell Specific ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human alpha Defensin 5 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is reactive with human samples and is run on a pre-coated 96-well microplate, read at 450 nm in a colorimetric microplate reader. The manufacturer states an assay time of 4h 30m, with kit components stored at +4°C or -20°C as instructed.

The assay has a detection range of 3.12-200 pg/ml and a sensitivity of 1.31 pg/ml. Standards are prepared as a dilution series from 200 pg/ml down to 3.13 pg/ml using the supplied Sample Dilution Buffer. The procedure uses a biotinylated anti-alpha Defensin 5 detection antibody followed by an HRP-avidin conjugate, with TMB substrate producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution; absorbance at 450 nm is proportional to analyte concentration.

Reported recovery values include serum at 80-102% (average 91%), EDTA plasma at 81-100% (average 90%) and heparin plasma at 80-89% (average 84%). Dilution linearity was demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma.
