From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of human Decorin in serum, plasma, tissue homogenates or other biological fluids. The kit employs a pre-coated microplate with an antibody specific for Decorin. Standards and samples are added to the wells and Decorin present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Decorin Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the Decorin concentration is calculated by reference to the standard curve.
The assay has a sensitivity of <35 pg/ml and a quantitative range of 78.1-5000 pg/ml, with a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date. Reported spike-and-recovery performance is 80-102% (average 91%) in serum, 81-100% (average 90%) in EDTA plasma, and 80-89% (average 84%) in heparin plasma, each evaluated with n=5 samples.
