A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Cystatin A in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is reactive with human samples and is read on a colorimetric microplate reader.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for Cystatin A is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, and any Cystatin A present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Cystatin A Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualize the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Cystatin A captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm against the standard curve. The platform is a pre-coated microplate (12 x 8 well strips) with an assay time of 4h 30m.

Reported performance includes a product range of 0.312–20 ng/ml and a sensitivity of <0.112 ng/ml. Recovery data: Serum (n=5) average 91%, range 80%–102%; EDTA Plasma (n=5) average 90%, range 81%–100%; Heparin Plasma (n=5) average 84%, range 80%–89%. Dilutional linearity was assessed for serum, EDTA plasma and heparin plasma samples tested at 1:2, 1:4, 1:8 and 1:16 dilutions (n=5 each). Kit components are to be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
