Human Cystatin 6 ELISA Kit

Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) for the in vitro quantitative determination of human CST6 (Cystatin 6) with reactivity against human samples. The kit is intended for serum, plasma, tissue homogenates, sweat, cell culture supernates and other biological fluids.

The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate. Standards and test samples are added to the wells, where CST6 is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-CST6 Antibody binds the captured analyte, an HRP-Avidin conjugate is added, and TMB substrate is used to visualise the enzymatic reaction, producing a blue colour that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the CST6 concentration is calculated against the standard curve. The standard curve spans a product range of 0.156–10 ng/ml with a sensitivity of <0.053 ng/ml. Total assay time is approximately 4 hours 30 minutes.

Reported spike recoveries (n=5) are 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma and 80–89% (average 84%) for heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 falls within 87–107% for serum, 79–108% for EDTA plasma and 82–105% for heparin plasma. Kit components should be stored at +4°C or –20°C as instructed and not used past the stated expiration date.
