Human Cylindromatosis ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human CYLD in tissue homogenates, cell lysates or other biological fluids, with reactivity for human samples. The assay employs the sandwich enzyme immunoassay technique: an antibody specific for CYLD is pre-coated onto a 96-well microplate, and standards or test samples are added so that the target is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-CYLD Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualize the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The absorbance is read at 450 nm in a microplate reader, and CYLD concentration is calculated against the standard curve.

The kit has a reported sensitivity of 0.117 ng/ml and an assay range of 0.312-20 ng/ml, with a total assay time of 4h 30m. Detection is colorimetric, performed on a pre-coated microplate comprising 12 x 8 well strips. Recovery has been evaluated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5) and heparin plasma (range 80%-89%, average 84%, n=5), with linearity of dilution assessed at 1:2, 1:4, 1:8 and 1:16 across the same sample types. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
