Human Cyclin Dependent Kinase 8 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of human Cdk8 (Cyclin Dependent Kinase 8) in tissue homogenates or other biological fluids. An antibody specific for Cdk8 is pre-coated onto a microplate; standards and samples are added and any Cdk8 present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Cdk8 Antibody is added, followed by an HRP-Avidin conjugate. The reaction is visualised using TMB substrate and the optical density read at 450 nm in a colorimetric microplate reader, with Cdk8 concentration calculated by reference to the standard curve.

The kit reports a sensitivity of <0.127 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Recovery in spiked samples averaged 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity was characterised at 1:2, 1:4, 1:8 and 1:16 across all three matrices. Supplied items include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers, with components stored at +4°C or −20°C as instructed.
