The assay uses a pre-coated 96-well microplate (12 x 8 well strips) bearing an immobilized antibody specific for Cdk6. Standards and samples are added to the wells, followed by a Biotinylated Anti-Cdk6 Antibody and an HRP-Avidin conjugate. A TMB substrate produces a colour change read at 450 nm in a colorimetric microplate reader, with Cdk6 concentration calculated by reference to a standard curve.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.053 ng/ml, and the total assay time is 4 hours 30 minutes.
