Human Cyclin Dependent Kinase 4 ELISA Kit

From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (ELISA) is designed for the in vitro quantitative determination of human Cyclin Dependent Kinase 4 (Cdk4) in tissue homogenates, cell lysates and other biological fluids. The assay is reactive with human samples and uses a pre-coated 96-well microplate arranged as 12 x 8 well strips.

The workflow employs an antibody specific for Cdk4 immobilised on the plate. Standards and test samples are added so that Cdk4 is captured in each well, followed by a biotinylated anti-Cdk4 antibody and an HRP-avidin conjugate. Bound HRP activity is visualised using TMB substrate, producing a blue colour that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of Cdk4 is calculated by comparison with the standard curve.

According to the manufacturer's specifications, the kit has a sensitivity of 0.121 ng/ml and an assay range of 0.312-20 ng/ml, with a total assay time of approximately 4 hours 30 minutes. In validation work using n=5 samples per matrix, recovery averaged 91% in serum (range 80%-102%), 90% in EDTA plasma (range 81%-100%) and 84% in heparin plasma (range 80%-89%). Kit components are stored at +4°C or -20°C as instructed, and the product should not be used past the stated expiration date.
