A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of human PCTAIRE1 (Cyclin Dependent Kinase 16) in tissue homogenates, cell lysates and other biological fluids. The assay uses a 96-well microplate pre-coated with antibody specific for PCTAIRE1. Standards and samples are added to the wells, where PCTAIRE1 is captured by the immobilised antibody. After washing, a Biotinylated Anti-PCTAIRE1 Antibody binds the captured analyte, followed by addition of an HRP-Avidin conjugate. TMB substrate is then added, producing a blue colour that converts to yellow upon addition of acidic stop solution, with the intensity of the yellow product proportional to the amount of PCTAIRE1 captured in each well.

Concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve. The reported assay range is 31.2-2000 pg/ml, with a sensitivity of <12.6 pg/ml. Total assay time is 4 h 30 m. Detection is colourimetric and the platform is a pre-coated microplate intended for use with a colorimetric microplate reader.

Reported mean recoveries in undiluted samples (n=5) are 91% in serum (range 83-100%), 96% in EDTA plasma (range 90-103%) and 95% in heparin plasma (range 89-102%). Dilution linearity data are provided for serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit must not be used past the expiration date.
