Human Complement Component 2 ELISA Kit

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of human Complement Component 2 (C2) in serum, plasma, and other biological fluids. The kit uses a pre-coated 96-well microplate (12 × 8 well strips) with an antibody specific for C2 immobilised on the wells. Standards or samples are added, and following incubation a biotinylated anti-C2 detection antibody binds the captured analyte. An HRP-avidin conjugate then catalyses TMB substrate, producing a blue product that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the C2 concentration is calculated against the standard curve.

The assay has a sensitivity of <0.224 ng/ml and a working range of 0.625–40 ng/ml. Spike-and-recovery in serum averages 91% (range 80%–102%, n=5), in EDTA plasma 90% (range 81%–99%, n=5), and in heparin plasma 84% (range 80%–89%, n=5). Dilutional linearity has been validated from 1:2 through 1:16 across serum, EDTA plasma and heparin plasma. Total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or −20°C as indicated in the manual, and the product should not be used past its expiration date.
