Human Complement Component 1, S Subcomponent ELISA Kit

Manufactured by Antibodies.com, this assay is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human Complement Component 1, S Subcomponent (C1s) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The plate is pre-coated with an antibody specific for C1s. Standards and test samples are incubated in the wells, followed by a Biotinylated Anti-C1s Antibody and an HRP-Avidin conjugate. A TMB substrate is then added; catalysis yields a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the C1s concentration is calculated against the standard curve.

The standard curve covers a working range of 31.2-2000 pg/ml with a sensitivity of <13.8 pg/ml. The platform is a pre-coated microplate arranged as 12 x 8 well strips, and the total assay time is approximately 4h 30m. Detection is colorimetric, performed on a standard microplate reader. Reported average recoveries from spiked samples are 91% for serum, 96% for EDTA plasma and 95% for heparin plasma.

Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored as instructed at +4°C or -20°C, and the kit should not be used past its expiration date.
