Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Complement Component 1, R Subcomponent (C1r) in serum, plasma or other biological fluids. The kit is reactive with human samples and is read on a colorimetric microplate reader at 450 nm.

The assay uses the sandwich enzyme immunoassay technique on a pre-coated microplate. An antibody specific for C1r is immobilised on the plate, standards and test samples are added, and bound C1r is detected with a biotinylated anti-C1r antibody followed by an HRP-avidin conjugate and TMB substrate. The density of the yellow product formed after addition of acidic stop solution is proportional to the amount of C1r captured, allowing quantification by reference to a standard curve. Total assay time is 4h 30m, and kit components are stored at +4°C or -20°C as instructed; the manufacturer advises not to use the kit past its expiration date.

Reported performance figures include a sensitivity of <0.061 ng/ml and an assay range of 0.156–10 ng/ml. Manufacturer validation data show recovery in serum (n=5) of 83–100% with an average of 91%, EDTA plasma (n=5) of 90–103% with an average of 96%, and heparin plasma (n=5) of 89–102% with an average of 95%. Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum (83–110%, 79–100%, 90–98%, 82–108%), EDTA plasma (90–98%, 90–105%, 73–109%, 87–91%) and heparin plasma (88–103%, 77–105%, 95–98%, 87–110%), each at n=5.
