Human Colony Stimulating Factor 2, Granulocyte Macrophage ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human GM-CSF (Granulocyte Macrophage Colony Stimulating Factor 2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for GM-CSF pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-GM-CSF Antibody and HRP-Avidin conjugate. Visualization is achieved using TMB substrate, which produces a blue-coloured product that turns yellow upon addition of acidic stop solution. The concentration of GM-CSF is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a product range of 15.6-1000 pg/ml and a sensitivity of <6.4 pg/ml. The total assay time is 4 hours 30 minutes, run on a pre-coated microplate of 12 x 8 well strips and read on a colorimetric microplate reader. Reported recovery averages are 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%) and 84% for heparin plasma (range 80%-89%). Dilution linearity (n=5) is reported across 1:2 to 1:16 dilutions for serum, EDTA plasma and heparin plasma.

Kit components supplied include a pre-coated 96 well microplate, lyophilized standard, detection solution A, detection solution B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers. Storage instructions call for components to be kept at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.
