Human Collagen Type XIV ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human COL14A1 in serum, plasma, tissue homogenates, cell culture supernatants, cell lysates and other biological fluids. The kit employs a 96-well microtiter plate pre-coated with an antibody specific for COL14A1, configured as 12 × 8 well strips. Standards and samples are added to the wells and bound by the immobilised antibody; after washing, a biotinylated anti-COL14A1 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader, with concentration calculated against the standard curve.

The kit has a reported sensitivity of 0.067 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Spiking recovery in serum averaged 91% (range 83%–100%), in EDTA plasma 96% (range 90%–103%), and in heparin plasma 95% (range 89%–102%). Linearity has been demonstrated across serial dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma. Components include lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at either +4°C or −20°C as instructed.
