Human Collagen Type X ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Collagen Type X in serum, plasma, tissue homogenates, and other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) with an immobilised antibody specific for Collagen Type X. Standards and test samples are added to the wells, where Collagen Type X is captured, followed by detection with a biotinylated anti-Collagen Type X antibody and an HRP-avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue colour that turns yellow upon addition of acidic stop solution, with absorbance measured at 450 nm on a colorimetric microplate reader.

The kit has a reported sensitivity of <0.141 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery has been characterised across sample types: serum (n=5) ranged from 83–100% with an average of 91%, EDTA plasma (n=5) from 90–103% with an average of 96%, and heparin plasma (n=5) from 89–102% with an average of 95%. Dilution linearity has been assessed at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma.

Kit components include the pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components should be stored at +4°C or –20°C as indicated in the product instructions, and the kit should not be used past its expiration date.
