Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human CD8 alpha in tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and employs an antibody specific for CD8 alpha, a biotinylated detection antibody, and an HRP-Avidin conjugate. Captured antigen is visualised with TMB substrate, producing a blue-coloured product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated against a standard curve.

The kit has a sensitivity of 1.3 ng/ml and a product range of 3.12-200 ng/ml. The complete assay runs in approximately 4 hours 30 minutes and is intended for research use only. Storage instructions specify holding kit components at +4°C or -20°C as indicated, and the product should not be used past its expiration date.

Manufacturer validation reports the following recovery ranges: serum 84%-102% (average 93%, n=5), EDTA plasma 91%-100% (average 95%, n=5), and heparin plasma 92%-98% (average 95%, n=5). Linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions: serum (n=5) at 90-103%, 77-108%, 86-93% and 91-106%; EDTA plasma (n=5) at 80-104%, 89-93%, 86-110% and 76-103%; heparin plasma (n=5) at 90-96%, 85-108%, 88-100% and 90-103%.
