Description

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of human CD19 in tissue homogenates, cell lysates, and other biological fluids. The assay uses a pre-coated microplate onto which an antibody specific for CD19 has been immobilised. Standards and test samples are added to the wells and any CD19 present is bound by the coated antibody. After incubation and washing, a biotinylated anti-CD19 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; the absorbance at 450 nm is read on a colorimetric microplate reader and the CD19 concentration is determined against the standard curve.

The assay has a reported sensitivity of 0.120 ng/ml and a standard curve range of 0.312–20 ng/ml. Validated sample types include tissue homogenates, cell lysates, serum, EDTA plasma, heparin plasma, cell culture supernatant, and other biological fluids. Reported average recovery values were 91% for serum (range 83–100%, n=5), 96% for EDTA plasma (range 90–103%, n=5), and 95% for heparin plasma (range 89–102%, n=5). Linearity data across 1:2 to 1:16 dilutions are also provided for serum, EDTA plasma, and heparin plasma. The total assay time is 4h 30m.

Required equipment not supplied with the kit includes a microplate reader capable of measuring absorbance at 450 nm ± 10 nm, a 37°C incubator, multi- and single-channel pipettes, and deionised or distilled water. Kit components are stored at +4°C or -20°C as instructed, and the product is intended for research use only.
