Human Cluster Of Differentiation 14 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Cluster Of Differentiation 14 (CD14) in serum, plasma, cell lysates or other biological fluids. The assay uses a 96-well microplate pre-coated with a CD14-specific antibody. Standards and samples are added to the wells, where CD14 is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-CD14 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Optical density is measured at 450 nm on a colorimetric microplate reader and CD14 concentration is calculated by reference to the standard curve.

Performance specifications include an assay range of 9.38-600 ng/ml and a sensitivity of <3.3 ng/ml. Total assay time is 4 hours 30 minutes. Kit components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Storage is at +4°C or -20°C as instructed; the kit should not be used past its expiration date.

Reported recovery averaged 91% in serum (range 83-100%, n=5), 96% in EDTA plasma (range 90-103%, n=5) and 95% in Heparin plasma (range 89-102%, n=5). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and Heparin plasma samples (n=5). The kit is suitable for serum, plasma and cell lysates.
