Human Claudin 3 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Claudin 3 in serum, plasma, tissue homogenates or other biological fluids. The kit uses a pre-coated 96-well microtiter plate supplied as 12 x 8 well strips, where standards and samples bind to a Claudin 3-specific capture antibody. After washing, a biotinylated anti-Claudin 3 detection antibody is added, followed by HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution; absorbance is read at 450 nm and the Claudin 3 concentration is calculated from the standard curve.

The assay has a sensitivity of <0.125 ng/ml and a quantitative range of 0.312–20 ng/ml, and is read on a colorimetric microplate reader. Total assay time is 4h 30m. Reported validation recovery data are: serum (n=5) 80%–102% with an average of 91%; EDTA plasma (n=5) 81%–100% with an average of 90%; and heparin plasma (n=5) 80%–89% with an average of 84%. Kit components are stored at +4°C or -20°C as instructed in the protocol and should not be used past the expiration date.
