Human Chromogranin B ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit is designed for the in vitro quantitative determination of human Chromogranin B in serum, plasma, tissue homogenates, and other biological fluids. The pre-coated microplate employs an antibody specific for Chromogranin B, with standards and samples added to the wells so that the analyte binds to the immobilized capture antibody. Following washing, a biotinylated anti-Chromogranin B antibody is added, then an HRP-Avidin conjugate, and the reaction is visualised with TMB substrate producing a blue product that turns yellow upon addition of stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of Chromogranin B is calculated against a standard curve.

The assay has a total run time of 4h 30m and a quantitative range of 62.5-4000 pg/ml, with a sensitivity of 22.5 pg/ml. Detection is colorimetric. Sample recovery has been validated across serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%), and heparin plasma (n=5, range 80%-89%, average 84%). Dilutional linearity was also assessed in serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16.

Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
